Dosage4 / 9
  • 100-300 mcg/kg
    • adult = 6-12 mg
  • cardiac standstill in NSX
    • onset 10-20 sec
    • 24 mg = 4 ampoules

Chemistry5 / 9

(molecular structure)

  • adenine attached to ribose
  • via beta-N9-glycosidic bond

Mechanism of action

(molecular effects)

Pharmacodynamics7 / 9

(therapeutic effects + indications / side effects + contraindications)

1:CVS

  • transient heart block at AV node
    • A1-receptor
    • inhibition adenylyl cyclase, ⬇cAMP, cell hyperpolarisation by ⬆K efflux, subsequent inhibition of Ca current
  • vasoD of normal segments of arteries
    • exaggerates the difference between normal and abnormal segments
    • reduces BQ distal to occlusion, causing ischaemia
    • coronary steal phenomenon
      • vasoD of healthy coronary As
      • diseased coronaries are already maximally dilated
      • this causes redistribution of BQ to non-ischaemic areas, causing myocardial ischaemia

2:RS

  • bronchospasm
    • CIN = asthma

3:NS

4:GIT

5:Renal

6:Endo

7:Haem

8:MSk
